Mi-sen Wu was born in Taipei, Taiwan. He received his BFA with Magna Cum Laude in film making from City College, City University of New York in 1995. His thesis film, Van Gogh's Ear which features the Beat Generation poet, Allen Ginsberg, won the most awards that the University has historically been conferred on any single film, including Best Film, Best Director, Best Cinematography and Best Editing. In the United States, it won additional honors from the Chicago Film Festival and New York Expo Showcase. Back at hometown, it also made a huge splash in Taiwan by winning the Best Film in the annual Golden Harvest Award. In 1998 the Taipei Film Festival chose only twenty short films over the entire history of Taiwan film production for exclusive screenings, to highlight the historical milestones. Among these twenty films were Ang Lee's Fine Line and Mi-sen Wu's Van Gogh's Ear.

In the year 2000, with the help of national film grant, Wu finished Fluffy Rhapsody, a narrative feature about a man who allows the tiny aches and pains of getting old and the fear of death transform his daily activities of job-searching. The film reveals a passion for the flesh from a very unconventional perspective. The film once again exhibits an assured sense of Wu's own unique visual style. Like his previous films, Fluffy Rhapsody has also been invited to many international film festivals, including Pusan International Film Festival, "New Currents" section, Taipei Golden Horse Film Festival, "New Chinese Beats" section, and Göteburg International Film Festival. The following year, it also won Wu another Best Film from Golden Harvest Award.

His first full-length feature, Drop Me a Cat, (2002) which depicts life in the desolation and emptiness of youth and examines the struggle to control the fate of millennium, won "Best Production Design" and received an nomination for "Best Music Score" from Golden Horse Film Awards. And his numerous other credits span the genres of documentary, experimental, animation, multimedia stage and TV commercial and music video. In 2006, Wu finished his second full-length feature film, Amour-Legende, (starring Yosuke Kubozuka), this labyrinthine, amnesiac drama was also funded by National grant, received Platinum Award from World-Fest Houston and "Best Sound Design" from Golden Horse Awards.
