And finally after 300 years, he met Aeng Cho through Hong Jo, and they fell in love all over again, with her forgiving him as well for her death.

To be honest, I really think it's become a Jang family thing to keep the wives off of the curse story. I initially thought that it's their patriarchal ways dominating considering that Sin Yu has also shielded Nayeon from it. That serious matters must be kept within the blood family aka the males haha.
But I also kinda thought that maybe it just became a custom to keep the women out of the curse specifics as who would marry them if they'd know about the curse, right? šš And then they just gotten used to not involving the women in these talks so the women really become oblivious about it like SinYu's mom.

I missed the question about Sin Yu's father.
It was explained that his father didn't die eventually because his disease was curable. It was then implied that all the other older Jangs' illnesses were curable but not Sin Yu's. That it all boils down to their illness management, but not for Sin Yu as his cannot be treated regardless. That also explains why the rest of the family is very protective of him, as well as why Sin Yu has chosen to keep his present condition secret from his family. He didn't want them to worry especially that he hasn't gotten married yet and has no offspring to leave behind. We can tell also that he's committed to his duty but for some reason he just couldn't love Nayeon enough to marry her right away.
Anyway, Sin Yu's sickness being incurable was such a puzzle to everyone until it was revealed that he was the reincarnation of Mu Jin. So his soul is basically the direct recipient of Aeng Cho's curse, of Aeng Cho's power, making his illness incurable. He's meant to die early like how Aeng Cho has meant as a punishment to whoever makes her heart stop beating.

Case in point was for the healing curse on Sin Yu. That was the only curse that was casted perfectly with the two of them present and without external pressure (Fair Skin spell was casted just to prove to Kwon Jaegyeong that spells worked). Sin Yu and Hong Jo were also very hopeful that it would work.
And then we eventually saw Sin Yu improving, that his body suddenly functions normally now and that the Red Hand no longer appeared.
But remember when Jaegyeong confronted their beliefs and ultimately made them believe that they were crazy to rely on the spells? Sin Yu could no longer see the effect of the Fair Skin spell on Hong Jo (his assistant suddenly mentioned that Hong Jo has gotten prettier), even Hong Jo (and Jaegyeong) herself couldn't see it. Plus Sin Yu's sickness crept back. He also began seeing the Red Hand again. (Others also thought that the Gardener's curse also had a hand on this, contradicting the healing spell but I also believe that it was a matter of Sin Yu no longer believing the powers of spells in general).
So what I'm trying to say is that the spells worked until they no longer believed it did.

The curse is related to the FL as she's the reincarnation of the person who casted it-- Aeng Cho. The curse cannot be broken by the FL. Aeng Cho never meant to have it be broken by her as she casted it in the first place to protect herself. Remember she suddenly just got baited into a trap which she didn't know who had set--- the concubine? the king? someone else? She also didn't expect that it would be the love of her life, Mu Jin, that would kill her and be cursed.
As Shaman Eun Wol has told Sin Yu "You killed the lady with the red hand. This is your karma." It is only the Jang descendant who could break it and it so happens that it was Sin Yu who broke it by dying for a bit before getting rescusitated.

The Jangs had more than one child but they happened to produce several males. If you remember the funeral that Sin Yu's family had, that was for his second cousin twice removed. And the details about the family having several sons (it's implied that they happen to produce sons only) can also be heard from the audio/gossips played while Sin Yu was entering the hall. And all of them have a short life span.
Yes, the Jangs knew that to stop the curse, they could also just die without producing an heir, but this contradicts their family duty as penned by Mu Jin to always have a male Jang to protect the shrine. If they die without an offspring, then they're in a way running away from their duty.
So to say, they took it upon themselves to just accept the curse just so they could fulfill the wishes of their ancestor. Remember how the Koreans are so bent on following family rules and traditions, almost to a fault. They believe in afterlife too that they're scared to meet their ancestors after death if they could not fulfill their duties while alive. Besides, they have a family shaman who somehow bridges the living and the dead for them so they know what a big sin it would be to go against their duty.
This is also why it was so rude of Hong Jo and the City of Onju to ask the Jangs to demolish the shrine as it meant so much to their family. They only accepted the demolition when it was Shaman EunWol herself that gave the green light whose powers and existence they so revered.
Also, the ending where Sin Yu's parents are having a girl is also implying that the family curse is indeed lifted as that the Jangs finally have a daughter for the first time.

And good point about mentioning SinYu's conversation with Shaman Eun Wol about him sharing that he's staying loyal with 2FL because in this particular scene, Eun Wol answers back with "Being loyal alone is the dumbest thing in the world".
This tells us that Eun Wol, being the powerful shaman that she is, can read through Sin Yu about Nayeon's infidelity and she's basically assuring him that it's okay to let go of Nayeon.
Remember this was a few days after Sin Yu apparently found out that Nayeon was cheating with him with Daum's CEO but his rational side tells him to stick with Nayeon after his confrontation with Hong Jo where the latter was obviously keen on denying her feelings for him. That she basically didn't want to get involved with Nayeon anymore, even if that includes Sin Yu. And even hurt him by saying that having feelings for him is undesirable.

His ancestors didn't break the curse because they died after having produced offspring/s so the latter already continued the curse. The curse was already passed to the next generation, the ancestor's death becomes irrelevant.
Basically, the male Jang descendants knew that they have a familial duty to protect the shrine and that they're cursed to die at a young age. For centuries, their only solution to perform their duty with the little life span they have is to immediately produce a male heir before they die. Sin Yu knew this too that's why he didn't really go against his betrothal to Nayeon. He was committed to perform his duty before his death (remember, he had a ticking time bomb).Who would have thought they could break off the curse if an heirless Jang male descendant die and then be rescusitated back to life?
